Varian TV 801 Turbo Pump Got You Down?
If you or your CRO partner have SCIEX API 4000™, 4000 QTRAP®, or API 5000™ systems, then you most likely are aware of the recent safety notice issued by SCIEX on the Varian TV 801 Turbo Pump. MicroConstants was not affected by this unfortunate pump recall for two reasons. First, we utilize both Waters and SCIEX systems and none of the Waters systems use this recalled Turbo Pump. As a precautionary measure to ensure that our sponsor’s projects are not subject to delay, we have duplicate instruments of the same model and have their crucial parts in supply. Although one of our Sciex API 4000 instruments was affected, we were able to switch out the defective pump with a replacement pump we had in stock. We had that instrument back up and running the same day the safety issue was received. Have a Humanized Antibody in the Preclinical Stages?
MicroConstants has developed a highly sensitive and specific immunoassay for the measurement of human IgG-based therapeutics, like antibodies and Fc-cytokine constructs, in preclinical specimens. The method involves Meso Scale Discovery (MSD®) electrochemiluminescence (ECL). Capture and detect antibody preparations were made from affinity-purified rabbit anti-human IgG that had been extensively adsorbed to eliminate non-human Ig reactivity. Wells of standard MSD plates are coated with anti-human IgG and, following incubations, washings, and sample additions, bound human IgG (or IgG-sonstruct) is detected with SULFO-TAG™-labeled rabbit anti-human IgG. The assay typically has a lower limit of quantitation (LLOQ) of 10-20 pg/mL in assay diluent, which translates to 1-2 ng/mL when accounting for the typical 1:100 minimum required dilution (MRD) of preclinical matrix. Cross-reactivity with IgG from other species (e.g., mouse, rat, monkey) is very low (less than 0.001%) allowing for the unequivocal identification of human IgG products in preclinical animal serum and plasma.
